Double-Sided Size Selection Method with NiXTips™
DPX Technologies has developed NiXTips™ for DNA clean up and size selection in Next Generation Sequencing library preparation that eliminates magnetic beads and magnets. NiXTips integrate proprietary silica media within a pipette tip to leverage aspirate-dispense steps to streamline DNA recovery while maximizing efficiency. This method demonstrates the capability of NiXTips for double-side size selection.
If the amplicon of interest is 240 bp and both smaller and larger fragments/amplicons must be removed, a two-sided size selection will be performed.
To begin, add binding buffer to the sample at a 0.5x ratio. Aspirate and dispense sample with a NiXTip for 30 cycles to bind the larger fragments to the right of the 0.5x curve (shaded area).
Add binding buffer to the sample with a transfer tip to reach a 0.8x ratio and mix. Use a second NiXTip to aspirate and dispense the sample 30 times to bind the fragments in the desired size range (shaded area). Wash NiXTips (other fragments left behind in well), then elute desired range.
