Automated Method for Improved DNA Isolation from Whole Blood with Novel Silica-based MicroPorous Xtraction (MPX) Technology

DNA isolation by silica binding is a robust and efficient technique used to recover high-quality genomic DNA from complex biological samples. Magnetic silica bead technology has been widely adopted for these extractions; however, bead capture time, bead carry over, bead resuspension and requirements for magnets on automation platforms complicate the process. DPX Technologies has developed a novel MicroPorous Xtraction Technology (MPX) that is incorporated into a pipette tip and eliminates magnetic beads, magnets for a fully automated, high throughput solution. This method uses the Hamilton robotics platform and a 1 mL MPX Tip from DPX Technologies for rapid bind, wash and elution steps. The MPX technology design and automated method minimizes cross-contamination and enhances reproducibility.

DNA extracted from 250 μL of whole blood using the 1 mL MPX Tip was compared to the leading bead-based and column-based technologies using their protocols. DNA yield (ng/µL) purity (260/280) and overall quality (gel electrophoresis) was compared. The MPX Tip method can process 96 samples from lysate to purified DNA in only 18 minutes and obtained higher yields of DNA and similar or better purities.

Innovative NIXTip™ Technology for Efficient DNA Fragment Capture and Size Selection in Sequencing Library Preparation

Solid Phase Reversible Immobilization (SPRI) with magnetic beads has been the stalwart for DNA clean up and size selection in Next Generation Sequencing protocols. The use of magnetic beads eliminates the need for centrifugation but has its own inherent draw backs such as bead capture time, bead carry over, bead suspension and required cumbersome magnets. DPX Technologies has developed NiXTip™ technology for DNA clean up and size selection in Next Generation Sequencing library preparation that eliminates magnetic beads and magnets. The NiXTip™ system utilizes a proprietary silica media integrated within a pipette tip to allow for aspirate-dispense steps that streamline DNA recovery while maximizing efficiency. Experimental procedures include DNA capture from PCR and enzymatic reaction cleanup, and double-sided size selection for sequencing libraries. Results demonstrate close to 100% recovery of DNA dependent on the target fragment size and buffer composition of the sample (different PCR master mixes). This data indicates that NiXTipTM technology can be used to simplify workflows, improve DNA yield, and provides an automated solution for processing up to 384 samples.
